An atomic force microscope (AFM) enables characterization of surfaces with nanometer resolution using a sharp tip mounted on a flexible cantilever that scans the surface of a sample. Interactions between the tip and the surface cause deflection of the cantilever, which is detected by a laser system, allowing determination of surface topography as well as mechanical properties. The system is coupled with a Raman microscope and also enables tip-enhanced Raman spectroscopy (TERS). In TERS, a metallic tip locally enhances the electromagnetic field and thus the Raman signal, enabling chemical analysis of surfaces with nanometer spatial resolution without the need for sample labeling.
