Content (Syllabus outline)

Development of plant tissue culture procedures. Short historical overview. Technical laboratory  demands for in vitro cultures. Conditions for optimal donor plant growth, organization of laboratory space, equippment ofr aseptic tissue manipulation, growth chambers, bioreactors.Media preprrations. Media constituents. Phytohormones: groups, influence in tissue culture and their effects.

Organogenesis: axillary and adventitious pathways. Stages of micropropagation.Specifics of direct organogenesis. Authotrophic micropropagation. Critical points of in vitro micropropagation: problems with axebic cultures and sporadic contaminations; hypersenzitivity; hyperhidration; shoot tip necrosis. Market oriented micropropagation including timing strategy.Examples of micropropagation.Long term line preservation methods. Slow in vitro growth, cryopreservation.Phytosanitary status of propagules.Indexing, detection of bacterial, fungal, viral and viroid infections.Test organisms, immunological methods,  rtPCR tests,  flow cytometric tests, microscopy. mikroskopija. Formation of virus-free seedlings.
